چکیده

Plants modify the polyunsaturated fatty acid content of their membrane and storage lipids in order to adapt to changes in temperature. In developing seeds, this response is largely controlled by the activities of the microsomal v-6 and v-3 fatty acid desaturases, FAD2 and FAD3. Although temperature regulation of desaturation has been studied at the molecular and biochemical levels, the genetic control of this trait is poorly understood. Here, we have characterized the response of Arabidopsis (Arabidopsis thaliana) seed lipids to variation in ambient temperature and found that heat inhibits both v-6 and v-3 desaturation in phosphatidylcholine, leading to a proportional change in triacylglycerol composition. Analysis of the 19 parental accessions of the multiparent advanced generation intercross (MAGIC) population showed that significant natural variation exists in the temperature responsiveness of v-6 desaturation. A combination of quantitative trait locus (QTL) analysis and genome-wide association studies (GWAS) using the MAGIC population suggests that v-6 desaturation is largely controlled by cis-acting sequence variants in the FAD2 59 untranslated region intron that determine the expression level of the gene. However, the temperature responsiveness of v-6 desaturation is controlled by a separate QTL on chromosome 2. The identity of this locus is unknown, but genome-wide association studies identified potentially causal sequence variants within ;40 genes in an ;450-kb region of the QTL.

Evaluation of the nutritive value of wild almond seed (Amygdalus scoparia) and its effect on performance, milk fatty acid composition and antioxidant activity in lactating goats

A total of 21 dairy goats in mid lactation were randomly assigned to three experimental diets in a completely randomized design. The diets contained 0, 3, or 6% WAS, and consisted of 45% forage and 55% concentrates. Milk fat and the ratio of milk fat to milk protein were significantly lower in goats fed diets containing 6% WAS (P≤0.01). Digestibility of crude fat was higher, and that of neutral...

Long-term administration of olanzapine induces adiposity and increases hepatic fatty acid desaturation protein in female C57BL/6J mice

Objective(s): Weight gain and metabolic disturbances such as dyslipidemia, are frequent side effects of second-generation antipsychotics, including olanzapine. This study examined the metabolic effects of chronic olanzapine exposure. In addition, we investigated the hepatic fatty acid effects of olanzapine in female C57BL/6J mice fed a normal diet.Materials and Methods: Female C57BL/6J mice ora...

Bioinformatics Genome-Wide Characterization of the WRKY Gene Family in Sorghum bicolor

The WRKY gene family encodes a large group of transcription factors that regulate genes involved in plant response to biotic and abiotic stresses. Sorghum is a notable grain and forage crop in semi-arid regions because of its unusual tolerance against hot and dry environments. We identified a set of 85 WRKY genes in the S. bicolor genome and classified them into three groups (I–III). Among the ...

Genome-wide identification and expression profiling of the fatty acid desaturase gene family in the silkworm, Bombyx mori.

Fatty acid desaturases exist in all living organisms and play important roles in many different biologic processes, such as fatty acid metabolism, lipid biosynthetic processes, and pheromone biosynthetic processes. Using the available silkworm genome sequence, we identified 14 candidate fatty acid desaturase genes. Eleven genes contain 3 conserved histidine cluster motifs and 4 transmembrane do...
